Triglyceride-glucose Index and Lung Cancer: Is There a Link?

  • The triglyceride-glucose (TyG) index level was not associated with an increased risk of lung cancer.

Why this matters

  • The exact mechanism between insulin resistance (IR) and lung cancer has not yet been fully understood; hence, further in-depth preclinical and clinical studies are warranted to determine the underlying pathophysiology.

Study design

  • An analysis of 324,334 participants (age, 40-70 years) without a history of any type of cancer, identified from the UK Biobank (2006-2010).

  • The association between IR measured by the TyG index and lung cancer risk was evaluated.

  • Interactions and joint effects of TyG index using a polygenic risk score (PRS) for lung cancer were also evaluated.

  • Funding: National Key Research and Development Program of China and others.

Key results

  • After adjustment for confounders, no significant association was observed between the TyG index and lung cancer risk (adjusted HR [aHR], 0.91; 95% CI, 0.64-1.18; P=.499).

  • After adjustment for PRS, there was no significant association between TyG index and lung cancer (aHR, 1.193; 95% CI, 0.901-1.580; P=.218).

  • Additionally, no significant interaction was observed between TyG index and lung cancer:

    • genetic score group (intermediate PRS): TyG index group (high), HR interaction, 0.767; 95% CI, 0.568-1.035; P=.083); and

    • genetic score group (high PRS): TyG index group (high), HR interaction, 0.835; 95% CI, 0.592-1.178; P=.304).


  • Study did not take into account physical activities and female reproductive factors.
